N. Stain (AFB) পরীক্ষা মূলত tuberculosis (TB) বা অন্যান্য acid-fast bacterial infections শনাক্ত করার জন্য করা হয়। এখানে AFB (Acid-Fast Bacilli), বিশেষ করে Mycobacterium tuberculosis, বিভিন্ন ধরনের clinical specimens থেকে খুঁজে বের করা হয়।
Z.N. stain বা Ziehl–Neelsen staining হলো একটি বিশেষ ধরণের staining technique যা bacteria কে red color এ প্রদর্শন করে এবং background কে blue/range করে। এই পরীক্ষার মাধ্যমে দ্রুত ধারণা পাওয়া যায় যে sample এ TB সংক্রমণ আছে কিনা।
এই পরীক্ষা সাধারণত নির্দেশ করা হয় যখন রোগীর মধ্যে persistent fever, weight loss, night sweats, cough (lung TB), lymph node swelling, or chronic wounds থাকে। Z.N. stain হলো rapid, preliminary diagnostic tool, তবে confirmatory diagnosis এর জন্য culture বা molecular test (PCR) প্রায়শই প্রয়োজন হয়।
Procedure: Sample (যেমন sputum, urine, wound swab, FNAC material) সংগ্রহের পরে smear তৈরি করা হয়। Smear Z.N. staining দ্বারা রঙিন করা হয় এবং microscope এ AFB presence পরীক্ষা করা হয়। AFB positive হলে Mycobacterium tuberculosis বা অন্যান্য acid-fast bacilli এর উপস্থিতি নির্দেশ করে।
Note: একবারের smear সবসময় নিশ্চিত ফলাফল দেয় না। সাধারণত multiple samples বা culture দিয়ে diagnosis নিশ্চিত করা হয়।
